Applications of the wide pore RP phases NUCLEODUR® 300

The NUCLEODUR® 300 RP phases are used for the analysis of biological macromolecules like proteins (s. appl. nos. 126580, 126800, 126570) and peptides (s. appl. no. 126610). Separations of protein and peptide mixtures with wide pore RP phases can be also used for the investigation of present substances and metabolites in biosynthesis and biochemical metabolism.

For the investigation of the tryptic digest of cytochrome c, being the sequence-specific cleavage by the serine protease trypsin, NUCLEODUR® 300-5 C18 features a better separation and less tailing than a competitor phase.

Cytochrome C types of different organisms can also be successfully separated with the MACHEREY-NAGEL wide pore RP 18 phase (s. appl. no. 126560). A fingerprint analysis with LC-MS of the tryptic digest of Cytochrome C (bos taurus) is shown with application no. 126820.


Wide pore NUCLEODUR® 300 RP phases expand the range of HPLC phases for separation of biological macromolcules. They are based on high purity silica of the latest generation. Due to a homogeneous coverage of the silica with C4 or C18 alkyl chains, high pH stability, LC-MS compatibility and high peak symmetries are possible for a reliable analysis, especially of proteins and peptides.
